Skin, Hyaluronic Acid and Collagen

Our skin is exceptionally alive, since its outer layer (the epidermis, rich in keratin) regenerates every 30 days or so.

The real metabolic centre of the skin is in the dermis (deep layer), rich in blood vessels and special cells (fibroblasts) that produce collagen, elastin and glycosaminoglycans (such as hyaluronic acid), giving the skin structure, hydration and resistance.

Collagen and Hyaluronic Acid

Collagen is an important structural protein, which plays a role in connective tissue to help maintain healthy skin function.

At skin level, collagen ensures the integrity, elasticity and "regeneration" of the skin. Collagen fibres provide scaffolding for elastin, which maintains skin elasticity, and for hyaluronic acid, which retains moisture and gives the skin moisture.

Collagen therefore helps to promote hydration, maintain elasticity and the firmness of the skin, reducing the presence of facial wrinkles.

Like hyaluronic acid and elastin, collagen is produced from connective tissue cells called fibroblasts. Together, these 3 substances contribute to the turgidity and firmness of young, healthy skin.

We could say, in fact, that the collagen fibres represent the backbone of our skin and give it resistance and tone, while the elastin keeps it elastic and the glycosaminoglycans ensure adequate hydration.

Wrinkles and Collagen

As the skin ages, the thickness of the extracellular matrix of the dermis (the structure that gives strength and resistance to the skin) decreases. As the solidity of this scaffold, mainly made up of collagen, disappears, the skin appears thinner and more wrinkled.

This phenomenon is caused by an imbalance between the production of collagen by fibroblasts and its degradation by lytic enzymes called metalloproteases. This is a normal process linked to the chronological ageing of the skin, which is significantly accelerated by excessive sun exposure.

The consequence of this degenerative phenomenon is a general disruption of the collagen fibres, which partially lose their supporting function causing a partial atrophy of the extracellular matrix, becoming soft and lacking in tone.

Over the course of a study (on a placebo, approved by an Ethics Committee, conducted in collaboration with Ambrosia Lab, a spin-off of the University of Ferrara), conducted on 30 female volunteers aged between 35 and 55 years:

• the application of the cosmetic X115^® alone for 8 weeks resulted in a 15% improvement in the collagen density of the treated area (the volar area of a forearm);
• the use of the X115^® supplement alone for 4 weeks resulted in an increase of 17%;
• the combined treatment: cosmetic for 8 weeks + supplement for 4 weeks produced improvements of 24%.

As an example, here are some images of the collagen density measured before and after treatment. The higher colour intensity indicates an increase in collagen density, while areas of low ecogenicity (black or green) indicate a more lax and less compact structure of the dermis, which is in fact more damaged. It is noted the significant increase in collagen density in all subjects treated and the absence of improvement in the control group not treated with X115^® (last image in the series)..

Skin ageing is an irreversible evolutionary process but fortunately it can be slowed down, even significantly, through the correct dermocosmetic and nutraceutical approach.

Among the various physiological alterations the skin suffers over the years, the loss of hydration contributes to the appearance of micro-wrinkles and the decrease in skin elasticity. This phenomenon is closely related to the composition of the fundamental substance, a gelatinous mass consisting of hyaluronic acid and proteoglycans, in turn consisting of glycosaminoglycans (long unbranched chains formed by disaccharide units) linked to protein chains.
